Born at Osho-ji in Kamo, Niigata, Yamaguchi hails from a family with a legacy in education and priesthood, notably her grandfather Nishimura Daikan, who founded Kamo Asa Gakuen and Kamo Aoi Kindergarten. She pursued her education at Seishin Joshi University in Tokyo after graduating from Sanjo High School in Niigata, where she immersed herself in acting through apprenticeships with theatrical groups En and Yumenoyasomusha.
Relocating to Hasuda, Saitama after marriage, she initially focused on homemaking but later entered politics at the encouragement of her father-in-law, a city councilor. Elected to the Hasuda City Council in 1999, she served five terms before successfully running as an independent candidate in the Saitama Prefectural Assembly election for Eastern District 5 in 2019, subsequently affiliating with the Liberal Democratic Party.
In 2022, Yamaguchi announced her candidacy for mayor of Hasuda, nominated by retiring mayor Kazunobu Nakano. Supported by the Liberal Democratic Party, Komeito party, and former Governor Motohiro Ono, she won the election on May 15, appealing to both conservative and independent voters with promises of local governance reforms.
