No, it's wordplay. The whole drama has lots of this between Su and JL. He has called her wife at least once using word play. Here the Emperor and Su were talking about pawns, but when the Emperor has confirmed that Su is in love with JL, his final words, "Take care of your pawn" becomes "Take care of your wife." Su smiled because he got the pun.

I think there are multiple meanings.
1) XFF suppressed herself through her previous life and even though she was the Cai Nue of her city (Most Talented Woman), everything she did was for her husband. She lost her Self. In this life as Jiang Li, she had been regaining her Self and Duke Su knew this. In the performance the previous night, he caught a glimpse of how his girl and her ex- used to be (following his lead) and he did not like what he saw. He told her in this life, she did not need to suppress herself like she did before.
2) It was also to inform her that he didn't see her as a pawn any more.
Maybe he wants to disrupt it from the inside? But you can't do that without first sleeping with the enemy, I guess?…
